The RAS/MAPK pathway (Ras-Raf-MEK-ERK signaling cascade) is involved in the regulation of cell proliferation, differentiation, and survival in response to extracellular signaling. BRAF is a member of the RAF family of protein kinases and represents one of the most frequently mutated protein kinase genes in human tumours. A mutation at nucleotide 1,799 results in the substitution of valine with glutamic acid at amino acid 600. Mutations in the braf gene is found in approximately 7%of cancers, and BRAF V600E accounts for more than 90% of these mutations.
