The epidermal growth factor receptor (EGFR) belongs to the ErbB family of receptor tyrosine kinases (RTKs). It is able to induce cell differentiation, proliferation, motility, and survival upon activation through binding of its ligands (e.g., EGF). Ligand binding triggers trans‐autophosphorylation of critical tyrosine residues in the receptor’s cytoplasmic tail and a subsequent signaling cascade mediated through interactions with downstream factors. Ligand‐induced EGFR phosphorylation and conformational change also lead to receptor endocytosis from the plasma membrane. EGFR is mutated and/or overexpressed in some human malignancies and is the target of various cancer therapies.
