LiveReceptor AMPAR

LiveReceptor AMPAR is the reagent which can specifically label AMPA-type glutamate receptors (AMPARs) with fluorescein dye on living neurons.
This reagent is based on a unique chemical reaction and can accomplish labeling of AMPARs keeping its ligand-binding pockets and receptor functions.
LiveReceptor AMPAR is a powerful tool to investigate physiological functions of AMPAR by monitoring receptor trafficking of endogenous AMPARs on live cells. No complicated protocol and genetic manipulation are required.

Generally, fluorescein direct conjugated ligand has been used for labeling receptors, however, such ligands easiliy detach from ligand binding pocket, and then signal will be faded out. (See below image).

 

 

“LiveReceptor” has a linker of acyl imidazole between fluorescein and ligand. While ligand is binding to receptor, nucleophillic amino acids attacks acyl imidazole and changes its chemical structure. By this method, receptors are directly labeled and keep fluorescence even after the ligand is detached from binding pocket.

 

 

This product is commercialized by the research result of Professor Hamachi, at Department of Synthetic Chemistry and Biological Chemistry, Graduate School of Engineering, Kyoto University.

  • Can label fluorecein to endogenous AMPAR

LipiDye II

LipiDye II is a high sensitive, low cytotoxic and super-photostable fluorescent dye for Lipid Droplets (LDs). LipiDye II enables to detect very small LDs (less than 1µm) and to perform long-term time-lapse imaging, including Z-stack imaging. This novel dye succeeded in observation of dynamic biosynthesis, degradation and movement of LDs in live cells.

  • High S/N ratio: Low background in cytoplasm via following two features

    2) Strong green fluorecent emission in non-polar oil phase like LDs

ULTRARIPA kit for Lipid Raft

PA buffer is one of the most useful buffers for protein extraction. RIPA buffer maintains most native structures of proteins and the extracted proteins can be applied to various applications. However, RIPA buffer is not sufficient to extract membrane proteins and membrane-associated proteins concentrated in lipid raft.

Lipid raft is a highly specialized microdomain on the lipid bilayer which contains specialized lipids, cholesterol and functional proteins. These lipid rafts are also called “Detergent Resistant Membrane (DRM)”, as lipid raft-enriched proteins are usually insoluble by mild detergent buffers such as 1% Triton X-100 and RIPA buffer. Consequently, it was difficult to analyze functions of lipid raft-enriched proteins extracted with RIPA buffer.

BioDynamics Laboratory’s newly developed product:the ULTRARIPA kit, can efficiently and rapidly extract membrane proteins or membrane-associated proteins enriched in lipid rafts, with little effect on protein structures and functions.

ULTRARIPA kit includes a totally new buffer not containing protein denaturing detergents, but can extract the DRM which was difficult to extract by conventional RIPA buffer. ULTRARIPA kit helps to analyze various biological assays of the proteins in lipid raft.
