Notes c-Myc proto-oncogene encodes nuclear DNA-binding phosphoproteins that are involved in the regulation of gene expression and DNA replication during cell growth and differentiation. Myc encodes a protein of 65 kDa which is expressed in almost all normal and transformed cells. The Myc protein activates transcription as part of a heteromeric complex with Max. Myc protein represses the expression of Ferritin-H, which sequesters intracellular iron, and stimulates the expression of IRP2 (Iron Regulatory Protein-2), which increases the intracellular iron pool. Myc induces transcription of the E2F1, E2F2 and E2F3 genes. Myc-induced S phase and apoptosis requires distinct E2F activities. The induction of specific E2F activities is an essential component in the Myc pathways that control cell proliferation and cell fate decisions.
